Hauptman"],"abstract":"Background: Medulloblastomas are the most common malignant brain tumors in children and are composed of distinct subtypes with different molecular origins. The SHH-subtype is associated with poorer prognoses and arises from granule neuron precursor cells, while the WNT-subtype presents later and has a better prognosis. Understanding these differences is crucial for diagnosis and treatment. Methods: The authors used gene expression databases to identify anatomical differences between SHH and WNT subtypes of medulloblastomas. They generated a mouse model with targeted deletion of the CTNNB1 gene in hindbrain cell populations to study its effects on tumor formation and cell proliferation. Results: The study found that SHH-subtype tumors were located in the cerebellum, while WNT-subtype tumors were found in the dorsal brainstem. CTNNB1 deletion led to abnormal cell proliferation in the dorsal brainstem but did not result in tumor formation unless paired with p53 loss, suggesting a cooperative role in WNT-subtype medulloblastomas. Conclusion: This research highlights the divergent molecular origins of medulloblastoma subtypes, which may significantly influence prognosis and therapeutic strategies. The findings suggest that future approaches to diagnosis and treatment should consider these distinct molecular characteristics.
